Functional relevance of the Solid-state photo-CIDNP effect

All of the natural photosynthetic reaction centers (RCs) we have studied, which originate from almost all photosynthetic families in the phylogenetic tree of life, exhibit the solid-state photo-CIDNP effect (Figure 1), although the window for the occurrence of the effect is quite narrow. The solid-state photo-CIDNP effect produces spin order. The effect seems to have been conserved in 3000000000 years of evolution. Therefore, we ask:

(i) What makes electron transfer in natural photosynthetic reaction centers (RCs) so efficient? Is the solid-state photo-CIDNP effect related to functional relevance?

(ii) How are cofactors such as chlorophylls tuned? What factors control electron transfer? What is the origin of the extremely high redox power of photosystem II that enables water oxidation?

(iii) What design principles of natural RCs can be adapted for the design of artificial photosynthesis? Do we need to implement the effect in artificial photosynthetic systems to make them more efficient?

New hyperpolarization methods in NMR and MRI

Magnetic-resonance methodology allows to study matter ‘from the inside’ in a non-destructive and non-invasive way. Hence, it is a perfect analytical approach. The traditional borders within magnetic-resonance methodology between the three methods called electron paramagnetic resonance (EPR), nuclear magnetic resonance (NMR) and magnetic resonance imaging (MRI), are presently lifted. Synergies can be realized as all methods rely on the same spin-physics. The methodology, deeply rooted in quantum mechanics, relies on weak magnetic interactions using low-energy micro- or radio-waves in the absence of ionizing radiation.

This unique analytical approach has been recognized by an unsurpassed number of Nobel prizes. The reason for this recognition is the versatility of the magnetic-resonance methodology: Applications are spanning the full range from fundamental physics, geo-science, materials science, analytical chemistry and biochemistry to structural biology as well as bio-medical and clinical purposes.

To overcome the intrinsically low Boltzmann ratio, limiting the signal intensity, we aim for pumping transient non-equilibrium polarization by means of physical processes and chemical reactions. This hyperpolarization can allow for signal enhancement of up to a factor of 100.000 opening new worlds of experiments. Figure 2 shows as example pumping of signals by the solid-state photo-CIDNP effect which might be transferred to MRI as contrast agent.

Spin-dynamics in electron-transfer dyads and proteins

Photosynthetic proteins and several flavoproteins (Figure 3) have been shown to convert light energy into transient nuclear hyperpolarization through spin evolution of spin-correlated radical pairs (SCRP). The light-induced hyperpolarization is also known as photochemically induced dynamic nuclear polarization (photo-CIDNP). Photo-CIDNP is observed in NMR spectroscopy under both liquid and solid-state conditions as non-Boltzmann nuclear magnetization, which allows a significant increase in signal intensity. The exact mechanism of photo-CIDNP formation, particularly in flavoproteins, is not yet fully understood.

Here, we aim to gain (i) insight into electron-electron nuclear dynamics and (ii) application as a tool for hyperpolarized experiments in NMR and MRI. To this end, we will mutate flavoproteins and study artificial dyads.

Functional principles of natural photoreceptors

Tetrapyrrole cofactors (also called bilins, see Figure 4) control numerous light-dependent processes in nature, such as photomorphogenesis, phototaxis or photoadaptation in plants, algae and bacteria. These proteins are often referred to as switches, ratchets, or molecular machines controlled by light.

The electronic structure expressed by color is related to the geometry, hydrogen bonding, and charge architecture of the surrounding protein. On the other hand, the light-induced change in the geometry of the open-chain tetrapyrrole cofactor is transferred to the surface of the protein.

The processes of cofactor-matrix interaction and signal transduction are often not yet understood. Solid-state and liquid-crystal NMR experiments reveal general design principles and provide ideas for synthesis and nanoscience.

Function and design of enzymes hydrolyzing polymers

Plastics have become an indispensable part of our daily lives due to their versatile applicability, leading to an annual increase in production. However, plastic waste can persist in nature for centuries without appropriate treatment, which causes a major environmental problem.

Polyethylene terephthalate (PET) is one of the most commonly used plastics, especially as packaging material for the food industry and as synthetic fibers for the textile industry. The biocatalytic degradation of PET by microbial enzymes has recently emerged as a promising option for a future eco-friendly recycling process for plastic waste. Unfortunately, biocatalytic recycling is not yet applied on an industrial scale due to the limited activity of previously known enzymes against post-consumer PET materials.

Therefore, we focus on studying structural and functional relationships of these enzymes using diverse biophysical and bioanalytical methods, in particular solution and solid-state NMR spectroscopy to provide an understanding of the enzymatic PET degradation mechanism by polyester hydrolases at the molecular level which would facilitate the engineering of more efficient enzyme variants.

Further projects

Micro-MRI resolves changes in organisms

The accumulation of microplastics in the environment is a problem that is receiving increasing attention. Little research has been done into the possible damage to living organisms caused by the absorption of microplastics. In an attempt to fill this gap, we will test the effect of PET (polyethylene terephthalate) microplastics on zebrafish (Danio rerio) embryos. Hence, we aim to study the fate of the microplastics in the organism, the toxicity of PET-microplastics in developing Zebrafish embryos and reconstruct the associated metabolic pathways.

To observe the transport of PET within the organism, we will apply fluorescence spectroscopy as well as 13C micro-magnetic resonance imaging (mMRI). The latter method also allows to study the chemical fate of the PET within an organism. High-resolution magic-angle spinning (HR-MAS) 1H NMR will allow for metabolic profiling based on statistical analyses as well as metabolite identification and quantification.

Since we want to compare effects of PET microplastics of different size and degree of crystallinity, the project includes PET synthesis, production of microplastics and polymer analytics including, e.g., Fourier-Transform Infrared (FT-IR) and solid-state NMR spectroscopy.
